Abstract: Alternative/modified methods for the analysis of Imidachloprid residue in/on banana were validated. Modifications were introduced in the first and second clean-up step based on a reference method (Method A) which used macroporous diatomaceous earth (MDE) column and prepacked Silica (Si) mini column respectively. The first modification (Method B) used liquid/liquid extraction as a substitute to MDE column in the first clean-up step; the second modification (Method C) used self-packed Si mini column as a substitute to prepacked Si mini column in the second clean up step; the third modification (Method D) used liquid/liquid extraction and self-packed Si mini column as a substitute to MDE column and prepacked Si mini column in the first and second clean up step respectively. The modified methods including the reference method showed satistaftory results per evaluation based on the parameters and method validation criteria. Method B showed comparable result with the reference method, based on DMRT, t-test and average percent recovery. Comparable result was likewise observed for Method D and the reference method. Method C which used self-packed Si mini column instead of prepacked Si mini column showed lower value in average percent recovery and significantly different from the other methods based on DMRT and t-test results. It can be inferred that packing technique of Si gel is very critical in the preparation of self-packed Si mini column. The study found that all the modified methods are applicable for the subsequent assay of Imidachloprid residue in/on banana. Method D is recommendable economic wise and in consideration of its high accuracy value in terms of percent recovery.
